Active Freeze Dryer Revolutionises Lyophilisation for Pharma

Innovative lyophilisation method simplifies validation for pharmaceutical manufacturers

An innovative lyophilisation method, known as Active Freeze Drying, is transforming the production of bulk pharmaceutical powders. Developed by Hosokawa Micron, this technology utilises a single, sterilisable, closed vessel to deliver fine, lump-free powder with significant advantages, including enhanced operator safety, reduced equipment requirements, and a contaminant-free final product. Validation is a critical challenge in pharmaceutical manufacturing, and freeze-drying is no exception. Manufacturers of active pharmaceutical ingredients (APIs) must comply with stringent standards set by regulatory bodies such as the EMA, FDA, cGMP, and GAMP. Single, Closed-Vessel System

Traditional tray/shelf freeze drying involves freezing material in individual vials on plates, making it difficult to ensure that all vials undergo the same process conditions, such as temperature and pressure. This approach often requires numerous temperature probes and time-consuming measurements.

In contrast, Hosokawa Micron’s Active Freeze Drying technology uses a single, closed vessel to aseptically freeze and dry the material. This method produces sterile bulk API, even in quantities as small as a few hundred grams. The closed-vessel design simplifies monitoring and ensures the homogeneity of the entire batch, significantly improving product quality.

The Active Freeze Drying system is capable of drying solutions, dispersions, pastes and wet solids. Successful applications include APIs, parenteral formulations, probiotics, and large-molecule products like PLGA and hyaluronic acid. The system features a jacketed and stirred dryer and a collection filter. The material is quickly frozen inside the vessel under agitation. The energy required for sublimation is applied through the jacket and efficiently distributed throughout the product by the stirrer, resulting in a fine, loose, free-flowing powder.

Unlike the hard, crusty material typically produced by tray drying, Active Freeze Drying technology yields a loose, free-flowing powder with a fine, uniform particle size and an open, porous structure. This eliminates the need for additional processing steps such as crushing, milling, or classification. The product’s porous nature facilitates effortless dissolution in water, making it ideal for applications like injectable solutions.

By avoiding a milling stage, the product remains undamaged, preserving the integrity of even the most delicate and sensitive structures, including living organisms. This gentle drying process ensures the final product maintains its quality, effectively achieving 'drying without harming'.

Aseptic Design

Hosokawa Micron's Active Freeze Drying system is meticulously designed for aseptic processing, ensuring product sterility throughout the entire process. The fully contained, single-vessel system eliminates contamination risks by maintaining a sterile environment from start to finish. The vessel's smooth, crevice-free design prevents bacterial growth, while sterile valves enable contamination-free material charging. After the drying cycle, the vacuum is broken using sterile gas, and the final dry powder is aseptically discharged into a sterilised container, which can be directly docked to a vial-filling machine. This process negates the need for additional containment equipment during the packing stage.

Cleaning and Sterilisation

The system's integrated Cleaning in Place (CIP) and Sterilisation in Place (SIP) processes make cleaning and sterilisation straightforward and highly effective. Spray nozzles inside the drying chamber initiate the cleaning process after each batch, with SIP achieved using pressurised steam or vapourised hydrogen peroxide. High heat transfer rates ensure rapid drying times, while easy filling, discharge, and quick cleaning cycles contribute to increased throughput. The fully contained design minimises manual intervention, eliminating the need for operatives in protective suits.

Compared to traditional tray freeze-drying, the Active Freeze Drying process is faster, more efficient, and significantly less labour-intensive. The system's efficiency allows for a single-operator workflow, streamlining the entire process. It produces lump-free, free-flowing powders in a single step, reducing handling times and costs. Additionally, its compact design conserves valuable floor space and reduces the need for additional equipment. This one-step approach saves pharmaceutical manufacturers time and money – an essential advantage in today's competitive market.

At the Hosokawa Micron Test Centre in the Netherlands, various unit sizes (1, 5, 60, and 800 litres) are available for customer testing to assess scalability and conduct trials.
